Molecular Signatures of Oxidative Stress Across Organ Systems: Implications for Rheumatoid Factor, Diabetes, and Hepatic Injury

ABSTRACT
Oxidative stress is a central molecular mechanism underlying diverse pathological conditions. It results from an imbalance between pro-oxidant species and antioxidant defenses, leading to widespread molecular damage via lipid peroxidation, protein oxidation, DNA damage, and perturbation of redox homeostasis. This review examines well-characterized molecular signatures of oxidative stress across organ systems, with emphasis on immune modulation, metabolic dysregulation, and liver pathology. We explore how reactive oxygen species (ROS) and related signaling pathways influence disease markers, including rheumatoid factor in autoimmune arthritis, impaired glucose metabolism in diabetes mellitus, and hepatocellular injury. The review synthesizes current understanding of key oxidative biomarkers (e.g., 8-oxo-2′-deoxyguanosine, malondialdehyde, protein carbonyls), transcriptional regulators (e.g., nuclear factor erythroid 2-related factor 2; Nrf2), inflammatory mediators (e.g., NF-κB), and mitochondrial dysfunction signatures across multiple tissues. We further discuss crosstalk between oxidative stress and immune function, metabolic regulation, and hepatic inflammatory cascades. Finally, we highlight translational implications for diagnostic strategies and antioxidant-based interventions. Understanding the molecular signatures common to these conditions may yield unified targets for therapeutic modulation of oxidative damage across organ systems.
